FAQs

What is the minimum genomic DNA (gDNA) concentration required for genotyping on the Biomark, Biomark HD or EP1 system? Why?

Fluidigm genotyping technology partitions each sample into multiple reaction chambers on the IFC. To avoid allele dropout, users should target 50 to 100 template copies (25 to 50 copies of each allele for a diploid organism) in every reaction chamber. The number of genomic copies in a given sample concentration will depend on the genome size and ploidy level of the genome of interest. For human samples, the genomic DNA concentration must be at least 60 ng/μL to attain this minimum copy number. The minimum concentration for other species should be considered relative to the human genome.
